НАН України. - 2003. - № 3. - С. 179-183. - Бібліогр.: 8 назв. - укp.It has been found recently that the platelet activating factor antagonist, ginkgolide B, is a selective use-dependent blacker of glycine-gated chloride channels. We have used various chloride channel blockers to get more insight into the mechanism of ginkgolide B blocking action. The <$E roman GABA sub A> receptor antagonist, picrotoxin, is known to block <$E alpha> homomeric glycine (Gly) receptors, while being ineffective for heteromeric <$E alpha sub 1 "/" beta> glycine receptors. Studying pyramidal hippocampal neurons of rats, we have found that the effect of picrotoxin depends on the age of the animals. Its blocking ability was characterized by <$E roman IC sub 50~=~140~mu>M and <$E roman IC sub 50~=~354~mu>M for 7- and 14-day old rats, respectively, indicating a possibly increased contribution of heteromeric receptors. We have found that the blocking action of ginkgolide B is subjected to a more drastic change in the same range of ages: the <$E roman IC sub 50> value is decreased from 1,6 <$E mu>M for 7-day old rats to 0,267 <$E mu>M for 14-day old rats. When measured against the background of ginkgolide B (1 <$E mu>M), <$E roman IC sub 50> for picrotoxin was 92 <$E mu>M. Taken together, these findings prompt that ginkgolide B might reveal more specific affinity to heteromeric Gly receptor-gated channels, than that for homomeric ones.
